Intact HIV genome quantification with 5-plex assay on Nio® Digital PCR
This application note presents the results obtained with the Nio®+ on reference samples using the Rainbow 5-plex assay.
A droplet can be positive for all five colors of the assay because it contains the intact genome fragment (all five targets present on the same DNA molecule), or because it co-encapsulates several smaller fragments such that all five targets are present but on several different DNA molecules.
To take into account the co-encapsulation events and accurately estimate the number of intact genome fragments,
mathematical calculations were used.

Highlights
- The all-in-one Nio®+ and its associated microfluidic consumable, the Ruby Chip, are suitable with dPCR high-order multiplexing approach such as color combination while significantly reducing the hands-on-time to results.
